      陈文连、男、1984年生人,农工民主党党员、研究员、上海中医药大学附属龙华医院基础研究部主任。

     (1)拓展“扶正治癌”学术思想之调控代谢重编程扶正的新内涵:健脾补肾是常用的扶正培本治法。我们的研究证实调控代谢重编程是健脾补肾法治疗肿瘤的现代医学新内涵。例如我们发现金复康发挥抗肿瘤活性的代谢靶点为:抑制外周血中的包括乙酰辅酶A在内的促癌代谢物、以及抑制肿瘤内ACLY介导的乙酰辅酶A和脂肪酸合成。

     (2)阐释健脾补肾法治疗肿瘤的物质基础及其作用新机制:发现补肾中药淫羊藿和女贞子共有的活性成分---齐墩果酸通过抑制嘌呤补救合成途径发挥抗肿瘤疗效。还发现益气健脾中药黄芪中黄酮类活性成分---异鼠李素通过抑制ACC1介导的脂肪酸合成发挥抗肿瘤疗效。

    (3)鉴定临床肿瘤患者的关键代谢通路并阐明其促癌机制,为中医药通过干预代谢重编程治疗肿瘤提供科学依据:创新性发现患者肿瘤细胞中存在一条新型能量代谢通路---GLUT5介导的果糖代谢,并阐明其促癌新机制:激活脂肪酸合成和AMPK/mTORC1信号。
